Smull, Christine E. (The Geisinger Medical Center, Danville, Pa.), and E. H. Ludwig. Infectivity of poliovirus and its nucleic acid for dehydrated HeLa cell monolayers. J. Bacteriol. 89:52-57. 1965.-A study was made of the infectivity of poliovirus ribonucleic acid (RNA), applied in various diluents, and of poliovirus, on cell monolayers which were washed free from their nutrient medium and allowed to dehydrate for periods up to 3 days prior to inoculation. The plaque formation of poliovirus RNA, applied in isotonic diluent, was greatly increased when assayed on cell monolayers dehydrated for certain periods of time. Conditions are described for bringing about optimal plaque formation by this means, and evidence is presented which strongly indicates that dehydration of the cell is an important factor in the increase in plaque formation of the poliovirus RNA. The plaque formation of poliovirus RNA in certain hypertonic diluents and in a basic protein-containing diluent was also improved with the use of dehydrated cell monolayers, whereas the plaque formation of poliovirus varied very little when assayed on cell monolayers dehydrated for periods up to 2 days. Under certain conditions, the size of the plaques increased as the dehydration time of the cell monolayers was increased. This was true with plaques initiated either by poliovirus or the nucleic acid. Further investigation in these cases revealed that virus production at given times was considerably greater in the dehydrated than in the undried cell monolayers.

Infectivity of Ribonucleic Acid from Poliovirus in Human Cell Monolayers
Ribonucleic acid prepared by the method of Gierer and Schramm from concentrated and partially purified types I and II polioviruses has been demonstrated to be infectious for HeLa and human amnion cells in monolayers. In areas of cytopathogenic action resulting from invasion of cells by RNA, intact poliovirus, of the type from which the RNA had been prepared, is present. The infectivity of the R...
متن کاملNucleic acid metabolism of virus-infected HeLa cells.
The study of the chemistry of virus-infected cells cannot yet claim a molecular interpretation for the genesis of the viral cytopathic effect. However, many striking observations have been made that must be accounted for in any molecular theory of viral cytopathology. From the viewpoint of comparative virology, the most consistent viral-induced aberrations involve the nucleic acids. The finding...
متن کاملIn-vitro assessment of the antiproliferative and apoptotic potential of the ethyl acetate extract of Peltophorum africanum on different cancer cell lines
We evaluated the in vitro antiproliferative and apoptotic potential of the ethyl acetate extract (EAE) of Peltophorum africanum, a member of the family Fabaceae (Sond) in order to validate its pharmacological use. Antiproliferation of human breast (MCF-7), colon (HT-29) and cervical (HeLa) cancer cell lines by EAE was investigated using the Cell Titer-Blue viability assay and the mechanism of a...

متن کاملDifferential inhibition of attachment and eclipse activities of HeLa cells for enteroviruses.
Receptor activities of HeLa cells were evaluated for ability to both attach and eclipse enteroviruses after exposure of cells to acid or heat. A modified procedure of acid (pH 1.5) elution of cell-associated virus, as compared with other procedures, provided a general method for the optimal recovery of receptor-bound enteroviruses. With this procedure, eclipse of virus operationally was conside...
